为提高纳豆芽孢杆菌对无机硒的生物转化率,通过单因素实验及响应面法对纳豆芽孢杆菌富硒的发酵条件进行优化,对样品进行透析及消化处理,并采用流动注射氢化物一火焰原子吸收光谱法测定有机硒含量,获得最佳有机硒转化率。优化后的发酵条件为:添加亚硒酸钠的同时接入菌株、亚硒酸钠质量浓度为10 μg/ml、培养基初始pH培养时间10 h、接种量为3.5%,最佳转化率达到91.21%。各单因素对有机硒转化率影响的大小为:接种量>培养基初始pH值>培养时间。

In order to improve the biotransformation rate of inorganic selenium by Bacillus natto, optimization of selenium-rich fermentation conditions of Bacillus natto by single-factor experiments and response surface methodology, treated sample by dialysis and digestion, the content of organic-se was determined by Flow injection-hydride generation-atomic absorption spectrometry to obtain the best organic selenium conversion rate. The optimized fermentation conditions are: the strain is added while adding sodium selenite, the mass concentration of sodium selenite is 10 μg/ml, the initial pH value is 7.5, the culture time is 10 hours, and the inoculation amount is 3.5%, The best conversion rate reached 91.21%. The results show that the effect of each single factor on the conversion rate of organic selenium is: inoculation amount> initial pH value > culture time
