摘要:
为了研究落葵种子色素提取过程中各种因素对于提取效果的影响及落葵种子色素的抑菌作用,本文通过单因素分析和正交实验设计的实验方法,以落葵果实种子干品为原料,将原料粉碎,采用溶剂浸提法提取,通过对不同提取条件下提取所得色素溶液在测定出的最适波长540nm下的比较各个条件下提取出溶液的吸光度来确定最佳浸落葵色素提液、最佳浸提时间、最佳浸提温度、浸提液料比、最佳浸提PH值以及最佳干燥方法。结果显示:落葵果实色素的最佳提取溶剂为水;30min为最佳浸提时间;25℃为最佳浸提温度;1:18为最佳液料比;PH值为5是最佳浸提PH值;水浴蒸干为最佳干燥方法。该提取条件成本低、选择性高、效果好,方便达成,所确定条件经济适用性高,快速方便。采用将浸满色素溶液的滤纸片放在涂布菌液的平板上培养观察是否存在抑菌圈的方法对落葵种子色素的抑菌作用进行了研究),实验菌种为金黄色葡萄球菌和大肠杆菌。结果显示:落葵果实色素对大肠杆菌和金黄色葡萄球菌无明显抑菌作用,对其它微生物的抑菌作用还有待于进一步研究。
Abstract:
In order to study the effect of various factors on the extraction of pigment from Basella alba seed and the bacteriostasis of seed pigment in Basella alba, this paper, by single factor analysis and orthogonal experimental design, uses the seed dry product of Basella alba as raw material to crush the raw material and extract it by solvent extraction, By comparing the absorbance of the pigment solution extracted under different extraction conditions at optimum wavelength 540nm, the optimum leaching solution, the best leaching time, the best leaching temperature, the leaching liquid material ratio, the best leaching ph value and the best drying method were determined. The results showed that the best extraction solvent for the fruit pigment was water, the best leaching time was 30min, the best leaching temperature was 25 ℃, the best liquid material ratio was 1:18, the best leaching ph was 5, and the best drying method was water bath evaporation. The extraction condition has the advantages of low cost, high selectivity, good effect, convenient achievement, high economic applicability and fast and convenient. The antibacterial activity of the pigment of the Basella alba seed was studied by using the filter paper which was soaked in the pigment solution on the flat plate of the coating bacterium liquid to observe whether the inhibition circle was present, and the experimental strains were Staphylococcus aureus and Escherichia coli. The results showed that the pigment had no obvious bacteriostasis effect on Escherichia coli and Staphylococcus aureus, and the bacteriostasis of other microorganisms had yet to be studied.
