以长裙竹荪抑菌物质的抑菌效果为指标 采用抑菌圈法筛选出水、90%乙醇、乙酸乙酯3种提取剂中的最佳提取剂,再以最佳提取剂提取长裙竹荪抑菌物质,对胡萝卜软腐欧文氏菌、层出镰刀霉两种果蔬致腐菌和大肠杆菌做抑菌敏感性实验,筛选出最敏感菌种。结果表明,乙酸乙酯提取的长裙竹荪浸提液的抑菌效果最好,而且乙酸乙酯为提取剂时,两种果蔬致腐菌均对长裙竹荪提取液表现出高度敏感,而大肠杆菌表现出低敏感度。最后以胡萝卜软腐欧文氏菌为目的菌,乙酸乙酯为提取剂,通过单因素和正交试验优化超声波辅助提取长裙竹荪抑菌物质的工艺。结果表明,各因素对长裙竹荪提取液抑菌效果的影响程度依次为料液比>超声时间>超声功率,最终优化提取条件为:固液比1:10,超声功率490 W,超声时间30 min。
Abstract:
Taking the bacteriostatic effect of bacteriostatic substances of Dictyophora longissima as the index, the best extraction agents were selected by the method of bacteriostatic circle. Then, the best extract was used to extract the bacteriostatic substances of Dictyophora longissima, and the most sensitive strains were screened through the bacteriostatic sensitivity experiments of Erwinia carotovora, Fusarium pullulans and E.coli. The results showed that ethyl acetate extract had the best bacteriostatic effect, and when ethyl acetate was used as the extract, the two kinds of fruit and vegetable rot causing bacteria were highly sensitive to the extract, while Escherichia coli showed low sensitivity. At last, the technology of ultrasonic assisted extraction of bacteriostatic substances from Dictyophora longissima was optimized by single factor and orthogonal test. The results showed that the ratio of material to liquid, ultrasonic time and ultrasonic power were the most important factors affecting the bacteriostatic effect of the extract. The optimal extraction conditions were solid-liquid ratio of 1:10, ultrasonic power of 490 W and ultrasonic time of 30 min.
