醋酸菌是一类专性好氧革兰氏阴性细菌,具有将乙醇氧化成乙酸的能力。以海藻酸钠(SA)和聚乙烯醇(PVA)两种高分子材料为载体对醋酸菌进行复合固定化研究,探讨PVA浓度、SA浓度和CaCl2浓度对醋酸菌固定化颗粒硬度、机械强度、包埋率以及产酸量的影响,优化复合固定化工艺条件。通过单因素实验和正交实验分析,结果表明,醋酸菌复合固定化最佳工艺条件为4%SA、10%PVA和0.6%CaCl2。在此条件下,固定化醋酸菌颗粒的硬度为2511.87±33.43 g、机械强度为100%、包埋率为86.09±0.81%,发酵产酸量为3.13±0.091 g/100 mL,较传统游离醋酸菌发酵提高了16.01%。
Abstract:
Acetic acid bacteria are a large group of obligate aerobic gram negative bacteria with the ability to oxidize ethanol to acetic acid. Sodium alginate (SA) and polyvinyl alcohol (PVA) were used as carriers to conduct composite immobilization of acetic acid bacteria. The effects of PVA concentration, SA concentration and CaCl2 concentration on the hardness, mechanical strength, embedding rate and acid yield of acetic acid bacteria immobilized particles were investigated to optimize the technological conditions of composite immobilization. Through single factor test and orthogonal test, the results showed that the optimal technological conditions for the composite immobilization of acetic acid bacteria were 4% SA,10% PVA and 0.6% CaCl2. Under these conditions, the hardness of immobilized acetic acid bacteria particles was 2511.87±33.43 g, the mechanical strength was 100%, the embedding rate was 86.09±0.81%, and the acid yield of fermentation was 3.13±0.091 g/100 mL, which was 16.01% higher than the traditional free acetic acid bacteria fermentation
