通过高效液相色谱图的分离效果、各个有机酸的线性相关关系、回收率及与常规方法作对比,确定SPE固相萃取柱、C18色谱柱,研究流动相种类、浓度及pH,检测条件的流速、温度、紫外检测波长的最优条件,以建立高效液相色谱法测定发酵液中7种有机酸的方法。结果表明:在通过SPE固相萃取柱和Tiank C18色谱柱进行分离后,选定了流动相为0.02moL/L(H3PO4(pH 2.7),流速为0.425mL/min,在紫外检测波长210 nm处,优化了测定条件。该方法回收率83%,相对标准偏差(RSD)<0.04%,有机酸的线性相关系数R2>0.994,可以实现7种有机酸的分离和准确定量。
Abstract:
The SPE solid phase extraction column and C18 chromatographic column were determined by comparing the separation effect of high performance liquid chromatography, the linear correlation of organic acids, the recovery rate and the conventional method. The optimum conditions of mobile phase type, concentration and pH, flow rate, temperature and ultraviolet detection wavelength of detection conditions were studied. A method for the determination of seven organic acids in zymotic fluid by high performance liquid chromatography was established. The results showed that the mobile phase was 0.02moL/L (H3PO4 (pH 2.7), the flow rate was 0.425mL/min, and the determination conditions were optimized at 210 nm of ultraviolet detection wavelength after separation by SPE solid phase extraction column and Tiank C18 column. The recovery rate of the method is 83%, the relative standard deviation (RSD) is less than 0.04%, and the linear correlation coefficient R2 of organic acids is more than 0.994. Seven kinds of organic acids can be separated and quantified accurately.
