本实验以耐寒性较好的东北分蘖洋葱为原料,利用冰结合磷酸缓冲溶液法,提取分蘖洋葱冰结构蛋白(Allium ice structural proteins,AISPs)。采用单因素和响应曲面法,考察了PBS溶液的浓度、料液比及冰球数量对冰结构蛋白提取率的影响。确定最佳参数为:分蘖洋葱滤汁与PBS溶液的体积比为1:5.32,PBS浓度为91.87 mmol/L,冰球添加量为33个/100 mL。在此条件下,AISPs的提取率达到65.23%。经全波长紫外扫描,在280 nm处有最大吸收峰。利用SDS-PAGE确定其分子量为:52 kDa,29 kDa,22 kDa,15 kDa。将AISPs添加到耐低糖酵母菌悬液中,当添加量为1.0 %、冻融周期为5d/次时,酵母菌的活菌数最高,菌落数为6.73×105个,存活率为51.45 %,分别比对照组提高了88.5 %和25.56 %。说明AISPs具有很好的抗冻保护活性。
Abstract:
In this experiment, the northeast tiller onion with good cold tolerance was used as raw material, and the ice-binding phosphate buffer solution method was used to extract the allium ice structural proteins(AISPs). Single factor and response surface method were used to investigate the effects of PBS solution concentration, solid-liquid ratio and hockey quantity on the extraction rate of ice structural protein. The optimal parameters were determined as follows: the volume ratio of tiller onion filtrate to PBS solution was 1:5.32, PBS concentration was 91.87 mmol/L, and the addition amount of hockey was 33 /100 mL. Under these conditions, the extraction rate of AISPs reached 65.23%. The maximum absorption peak at 280 nm was obtained by full wavelength uv scanning. The molecular weight was determined by sds-page as 52 kDa, 29 kDa, 22 kDa and 15 kDa. When AISPs were added to the suspension of low-glycus-tolerant yeast, when the addition amount was 1.0 % and the freeze-thaw cycle was 5d/ time, the number of viable bacteria in yeast was the highest, with the number of colonies being 6.73×105 and the survival rate being 51.45%, which was 88.5 % and 25.56 % higher than the control group, respectively. These results indicate that AISPs have good antifreeze protection activity
