榨菜作为一种发酵食品,它的制作过程中酵母菌发挥了不可或缺的作用,为了研究榨菜中酵母菌的多样性,本实验以市售爽口榨菜和乌江榨菜为原材料,采用分子生物学方法PCR—DGGE的非培养方法进行测定,通过变性梯度凝胶电泳DGGE技术把聚合酶链式反应PCR扩增出来的DNA进行分离,从而得到不同的基因序列;通过UPGAME算法对榨菜中酵母菌进行相似性聚类分析。结果为经序列分析可得爽口榨菜(SZ)中有两种不同序列的酵母菌,分别与Candida tropicalis、Pichia fermentans这两种酵母菌的同源性为95%、88%,而在乌江榨菜(WZ)中也有两种不同序列的酵母菌,WZ中的第一条条带的变性浓度与SZ中第一条条带相同,故分析可知为同一种酵母菌,WZ中的第二条条带的酵母菌与Saturnispora mendonce的同源性为95%。通过Microsoft Excel软件计算其多样性指数、丰富度、均匀度可知,爽口榨菜(SZ)中的酵母菌多样性指数与丰富度都比乌江榨菜(WZ)中的数值大,但均匀度却比WZ小
Abstract:
ABSTRACT: Pickle as a kind of fermented foods, saccharomycetes played an indispensable role in the making process of the preserved szechuan pickle , in order to study the diversity of the saccharomycetes in pickle, this experiment is refreshing pickle and wujiang river pickle as raw material, using PCR - DGGE molecular biology method of cultivation methods were determined, by denaturing gradient gel electrophoresis DGGE technology segments of the polymerase chain reaction PCR amplification of the same size but different sequences of DNA separation, resulting in different species of saccharomycetes ; Through the Quantity One similarity clustering analysis software. Sequence analysis results for the available and refreshing pickle (SZ) there are two different sequence of saccharomycetes, respectively with Candida tropicalis, Pichia fermentans both saccharomycetes homology of 95%, 88%, and in the wujiang river pickle (WZ) also have two different sequence of saccharomycetes, the first transgender concentrations of the stripes in WZ same as the first stripes in SZ, so the analysis for the same kind of saccharomycetes, second of WZ all with saccharomycetes and Saturnispora mendonce homology of 95%. By Microsoft Excel software to calculate the diversity index, richness and evenness, and refreshing pickle (SZ) of saccharomycetes diversity index and richness is greater than the value in wujiang river pickle (WZ), but smaller than WZ evenness.
