本文采用超声辅助法对富硒黑木耳多糖和普通黑木耳多糖进行提取和分离纯化,得到富硒黑木耳多糖(Se-AAP-2)和普通黑木耳多糖(AAP-2),比较分析Se-AAP-2和AAP-2的分子量、单糖组成、纯度、糖醛酸含量、体外及体内抗氧化活性。结果表明:Se-AAP-2和AAP-2均由甘露糖和葡萄糖组成,分子摩尔比分别为1:1.31和1:1.28;相比于AAP-2,Se-AAP-2平均分子量降低了23.5%(P<0.01),糖醛酸含量提高了33.7%(P<0.01),说明富硒能够显著降低黑木耳多糖的分子量,提升其糖醛酸含量。此外,Se-AAP-2拥有更高的体外的抗氧化活性,相比APP-2,其总还原力提高了11.4%,羟基自由基以及ABTS自由基清除能力IC50亦分别降低了42.2%和26.4%。体内抗氧化活性评价结果表明,Se-AAP-2相较于AAP-2具有更强的体内抗氧化活性,能够显著提高小鼠肝、肾、心以及血清中T-SOD、GSH-Px的活性,降低其MDA含量。

The study used ultrasound-assisted method to extract, separate and purify selenium-enriched Auricularia auricula polysaccharides and common Auricularia auricula polysaccharides to obtain selenium-enriched Auricularia auricula polysaccharides (Se-AAP-2) and common Auricularia auricula polysaccharide (AAP-2). The molecular weight, monosaccharide composition, purity, uronic acid content, and antioxidant activity of Se-AAP-2 and AAP-2 were compared and analyzed. The results showed that Se-AAP-2 and AAP-2 were both composed of mannose and glucose, and the molecular molar ratios were 1:1.31 and 1: 1.28 respectively; compared with AAP-2, the average molecular weight of Se-AAP-2 was reduced by 23.5% (P<0.01), and the uronic acid content was increased by 33.7% (P<0.01), indicating that selenium enrichment could significantly reduce the molecular weight of Auricularia auricula polysaccharides and increase its uronic acid content. In addition, Se-AAP-2 had higher in vitro antioxidant activity. Compared with AAP-2, its total reducing power was increased by 11.4%, and the ?OH and ABTS free radical scavenging ability IC50 was also reduced by 42.2% and 26.4%. The evaluation results of in vivo antioxidant activity showed that Se-AAP-2 had stronger in vivo antioxidant activity than AAP-2, it could significantly increase the activity of T-SOD and GSH-Px in the liver, kidney, heart and serum of mice and reduce its MDA content.
