为研究仙草黄酮纯化工艺及其主要组分的结构,以仙草为原料,采用大孔树脂(HPD-100)、羟丙基葡聚糖凝胶(Sephadex LH-20)分别对仙草黄酮进行柱层析纯化和组分分离,利用HPLC、1H-NMR、13C-NMR对分离得到的化合物进行分析和结构鉴定。结果表明,HPD-100大孔树脂纯化仙草黄酮效果良好,动态吸附工艺参数为:上样量为3 BV,上样流速为3 BV/h,洗脱剂乙醇浓度为60 %,洗脱流速为6 BV /h。通过Sephadex LH-20柱层析分离得到两种黄酮类化合物,HPLC分析表明两者纯度皆在90 %以上,经鉴定为迷迭香酸(rosmarinic acid,MBF1)与紫云英苷(astragaloside,MBF2)。本研究可为仙草资源的高值化利用提供一定的理论依据。

In order to study the purification process and main components of Flavonoids from the Mesona Blume., the constituents of the extract were separated and purified by macroporous resin (HPD-100) and hydroxypropyl dextran gel (Sephadex LH-20). The main components were separated and analyzed by HPLC, 1H-NMR and 13C-NMR. The results showed that HPD-100 macroporous resin had a good purification effect. The dynamic adsorption parameters were as follows: the amount of sample was 3 BV, the flow rate of sample was 3 bv / h, the concentration of eluent ethanol was 60 %, and the flow rate of eluent was 6 bv / h. Two flavonoids were separated by Sephadex LH-20 column chromatography. HPLC analysis showed that the purity of both flavonoids was over 90 %. They were identified as rosmarinic acid (MBF1) and astragaloside (MBF2). This study can provide a theoretical basis for the high value utilization of Mesona Blume.o resources.
