探讨无花果多糖分离纯化工艺及抗氧化性能,以无花果为试材,通过测定无花果中的使用AB-8树脂去除多糖中的色素,利用3%三氯乙酸法去除蛋白质,选用0.03 mg/mL的?NaCL溶液洗脱纯化,使用AB-8柱层析色谱图在管数为10-20、30-40中出现脱峰分离纯化完成。通过测定清除超氧阴离子自由基、羟基自由基和还原能力,评价其抗氧化性能。结果表明,当浓度为2500μg/mL时,无花果超氧阴离子效率和羟基自由基清除效率都达到最高,分别为75%和38%。在1000μg/mL浓度下,还原量最大,无花果多糖分离纯化后具有较高的抗氧化性能,研究结果可为无花果产业生产提供技术支持。
Abstract:
To explore the separation and purification process and antioxidant properties of polysaccharides from figs. Figs were used as test materials to remove pigments from polysaccharides by measuring AB-8 resin in figs, and to remove proteins by 3% trichloroacetic acid method, using 0.03 mg/mL The NaCL solution was eluted and purified, and the AB-8 column chromatogram was used to show off peak separation and purification in the tubes of 10-20 and 30-40. By measuring the scavenging capacity of superoxide anion free radicals, hydroxyl free radicals and reducing, the antioxidant performance was evaluated. The results showed that when the concentration was 2500μg/mL, the superoxide anion efficiency and hydroxyl radical scavenging efficiency of fig reached the highest, which were 75% and 38%, respectively. At a concentration of 1000μg/mL, the reduction amount is the largest, and the fig polysaccharide has high antioxidant performance after separation and purification. The research results can provide technical support for the production of fig industry.
