为优化红蓝草红色素的最佳提取工艺,研究其稳定性及抗氧化活性。本试验以广西红蓝草为原料,吸光值为指标,在单因素试验的基础上,通过响应面分析模型对红蓝草红色素提取工艺条件进行优化分析,研究温度、pH、蔗糖、食盐、还原剂、防腐剂及金属离子对其稳定性的影响,通过测定红蓝草红色素对·OH、O2-和DPPH·的清除能力以评价其抗氧化活性。结果表明:红蓝草红色素最佳提取方法为超声波辅助水提法,最优提取条件为料液比1:25 g/mL,超声温度53 ℃,超声时间109 min,超声功率160 W;红蓝草红色素对温度、蔗糖、食盐、防腐剂和大部分金属离子具有较好的耐受性,在中性偏酸条件下稳定,在碱性条件中则易降解,其中高浓度蔗糖、食盐溶液、Al3+及还原剂对其起增色作用;通过测定红蓝草红色素对O2-、·OH、DPPH·的清除能力,得出红蓝草红色素具有较高抗氧化活性和稳定性。
Abstract:
In order to optimize the optimal extraction process of Spodiopogon cotulifer’s red pigment and study its stability and antioxidant activity, based on the single factor test, the extraction process conditions of Spodiopogon cotulifer’s red pigment were optimized through the response surface analysis model. Guangxi Spodiopogon cotulifer was used as the raw material, and the absorbance value was used as an indicator. At the same time, the effects of temperature, pH, sucrose, common salt, the reducing agents preservatives and metal ions on their stability were also studied. Their antioxidant capacity was evaluated by measuring the scavenging ability of Spodiopogon cotulifer’s red pigment to ·OH, O2-, and DPPH· active. The results showed that the preferred extraction method of Spodiopogon cotulifer’s red pigment was ultrasonic-assisted water extraction. The optimal extraction conditions were a material-liquid ratio of 1:25, an ultrasonic temperature of 53 ℃, an ultrasonic time of 109 minutes, and an ultrasonic power of 160 W. Spodiopogon cotulifer’s red pigment had good tolerance to temperature, sucrose, common salt, preservatives and most metal ions. It was stable in a neutral acidic conditions and can be easily degraded in an alkaline conditions. High-concentration sucrose, common salt solution, Al3+, and the reducing agent can enhance its color. By measuring the scavenging ability of Spodiopogon cotulifer’s red pigment to O2-, ·OH and DPPH·, it was concluded that Spodiopogon cotulifer’s red pigment had high antioxidant activity and stability.
