建立了高效液相色谱法测定大蒜愈伤组织中大蒜素含量的分析方法。色谱柱Eclipse Plus C18,5μm,4.6×250 mm,柱温35℃,流动相为甲醇-水含0.2%甲酸(80:20),流速1.0 ml/min,二极管阵列检测器检测波长240 nm,进样量50 μL。结果显示,大蒜素在0.30~4.57 μg/ml范围内线性关系良好,相关系数R2=0.999,加标回收率为95% ~ 102%。该方法灵敏度高,重现性好,操作简便,适用于大蒜愈伤组织中大蒜素含量的检测。
Abstract:
Develop a method for the determination of allicin in garlic callus by high performance liquid chromatography (HPLC). The conditions of HPLC are taken as follows: the chromatography column is Eclipse Plus C18(5μm,4.6×250 mm℃, the mobile phase methanol : water (0.2 formic acid) is 80:20(V/V), the flow rate is 1.0 ml/min, the DAD detector wavelength is 240 nm, and the sample injection volume is 50 μL. The results show that the content of allicin is in the range of 0.30 ~ 4.57 μg/ml, it has a good linear relationship, and the correlation coefficient is 0.999, and the average recovery of allicin is 95% ~ 102%. This method is accurate, reliable, simple and efficient for the detection of allicin in garlic callus
